Research Report Summary

Herbal Formula Chai Hu Jia Long Gu Mu Li Tang,  Ameliorates Depression through Modulating Hypothalamo-Pituitary-Adrenal (HPA) Axis

A classical Chinese herbal formula, Chai Hu Jia Long Gu Mu Li Tang or Saiko-ka-ryukotsu-borei-to in (kanpo) Japanese herbal medicine, has been used to treat depression, anxiety and insomnia. About one half of depression is caused by attenuation of the dexamethasone-mediated negative feedback on cortisol secretion, which results in disruption of hypothalamo-pituitary-adrenal (HPA) axis.

The researchers at Japan's National Institute for Longevity Sciences examined the effects of Chai Hu Jia Long Gu Mu Li Tang on the chronic stress-induced changes in glucocorticoid receptor (GR) in the PFC and hippocampus. Chronic stress was induced in rats by water immersion and restraint (2 h/day) for 4 weeks.  Chai Hu Jia Long Gu Mu Li Tang significantly prevented decreased cytosolic GRs in the PFC and increased nuclear GRs in the hippocampus in the chronically stressed rats. Moreover, Chai Hu Jia Long Gu Mu Li Tang significantly prevented the abolishment of feedback ability in both regions of brain. These results suggest that the beneficial effects of Chai Hu Jia Long Gu Mu Li Tang on the glucocorticoid receptor level are involved in its ameliorating actions on the HPA disruption and depression.
